ABSTRACT

Objective:To explore the potential mechanism of Fuzheng Jiedu Decoction created by professor Yu Huiping in the treatment of primary immune thrombocytopenia (ITP) in children based on network pharmacology.Methods:The targets of Fuzheng Jiedu Decoction and ITP were retrieved within SymMap database and TCMID database, and all the common genes in the potential targets of the decoction and ITP were retained. The interaction relationship among the targets was obtained in the String database, and cluster analysis was conducted to obtain the core target group of Fuzheng Jiedu Decoction for ITP. In the David database, the potential KEGG Pathway was obtained through enrichment analysis, the Pathway of non-specific diseases was classified and selected, and a network of "Traditional Chinese Medicine - Target - Pathway" was constructed.Results:There are 500 potential targets for Fuzheng Jiedu Decoction to treat ITP. After Cluster analysis of PPI network, a total of 16 gene clusters were obtained, among which Cluster 1 score was 65.663, making it a potential core target group for Fuzheng Jiedu Decoction to treat ITP. The core enriched target group amounts to 114 pathways, and there were four first-level catalogs which includes Human Diseases (50%), Organismal Systems (25%), Environmental Information Processing (17%), and Cellular Processes (8%). Among them, TNF signaling pathway and HIF-1 signaling pathway were highly enriched for non-specific diseases. In the nodes of the network, The Chinese herbs with the highest Degree of aggregation in the network nodes were Agrimoniae herba (Degree=66), Glycyrrhizae radix et rhizoma praeparata cum melle (Degree=64), the target proteins were MAPK3 (Degree=51),MAPK1 (Degree=50),and the pathway was PI3K-Akt signaling pathway (Degree=29). Conclusion:Fuzheng Jiedu Decoction is mainly used to treat children's ITP with Agrimoniae herba and Glycyrrhizae radix et rhizoma praeparata cum melle,and it is related to the regulation of platelet number, adhesion and focusing.

ABSTRACT

Objective To investigate the clinicopathological characteristics and the expression of related molecular markers of heterotopic pancreas for better understanding and avoiding overtreatment of this disease.Methods From 24th March 2009 to 10th November 2016,92 patients with heterotopic pancreas in upper digestive tract diagnosed after endoscopic submucosal dissection(ESD),were collected. Tissues were sectioned and pathologically classified by Heinrich classification.The expressions of seven different molecular markers including cytokeratin(CK)19,insulin,trypsin,Ki-67,p53,CD133 and CD56 were detected by immunohistochemistry staining. Clinical features, pathological features and immunohistochemical results were retrospectively analyzed.Analysis of variance and Kruskal-Wallis test were used.Results According to Heinrich classification,the percentages of type Ⅰ,Ⅱ,and Ⅲ of heterotopic pancreas were 27.2%(25/92),63.0%(58/92)and 9.8%(9/92),respectively.There was no type Ⅳ patients.The heterotopic pancreas mainly located in stomach with proportion being about 91.3%(84/92)of the total heterotopic pancreas.CK19(the marker of pancreas ducts),insulin(as marker of endocrine function)and trypsin(as the marker of exocrine function)were all expressed in heterotopic pancreas.The positive rate of CD56,a pancreatic neuroendocrine marker,was 66.3%(61/92).The umbilicus like depression was the typical endoscopic appearance of heterotopic pancreas,which only found in 29 patients(31.5%).The average rate of Ki-67,cell proliferation index,was(2.08 ± 1.41)%.The expression of mutant p53 was negative in all 92 cases of heterotopic pancreas.The average staining area of CD133,a marker of pancreatic cancer stem cell,was(2.53 ± 2.43)%.The average follow-up period of 92 patients was(43.6 ± 27.5)months.No relapse and malignant change were found and all patients survived after ESD.Conclusions Heterotopic pancreas has normal pancreatic construction and function.The cell proliferation index is low in heterotopic pancreas and no mutant p53 expression is found.The expression of CD133 is also low.Heterotopic pancreas is a congenital benign disease which requires a long-term follow-up rather than overtreatment.

ABSTRACT

Objective To investigate the effect of metoclopramide combined with psychological intervention on nausea and vomiting after laparoscopic cholecystectomy (LC). Methods From January 2015 to January 2017, 90 patients with nausea and vomiting after laparoscopic cholecystectomy according to the different treatment and nursing intervention mode were divided into the control group and the observation group, 45cases in each group. The control group were given granisetron and routine nursing, the observation group were received metoclopramide and psychological intervention. The experimental data was recorded and compared, the improvement of nausea and vomiting were observed. Results The clinical effect in the observation group is better than that in the control group. The improvement of nausea and vomiting, the nursing satisfaction in the observation group were better than those in the control group. The differences were statistically significant (P<0.05). Conclusion Metoclopramide combined with psychological intervention on the treatment of the patients with nausea and vomiting after laparoscopic cholecystectomy, curative effect is obvious, which can effectively improve the nausea and vomiting, nursing satisfaction is high, which is worthy of clinical application.

ABSTRACT

Objective To observe the real-time efficacy of balance acupuncture in treating primary hypertension. Method Totally 160 patients with primary hypertension were randomized into a treatment group and a control group, 80 in each group. The treatment group was intervened by balance acupuncture, while the control group was by oral administration of Captopril. The blood pressure and symptom score were observed before and after intervention. Result There were no significant differences in comparing the total effective rate at different time points between the two groups (P>0.05). There was a significant difference in comparing the headache score in male patients at 30 min after treatment between the two groups (P<0.05). Conclusion Balance acupuncture is an effective method in treating primary hypertension, and can release headache of the patients.

ABSTRACT

Objective:To investigate the expression of Bcl-2 in rat brain during dependence and withdral state.Methods:The rats were rendered dependent by subcutaneous injections of increasing doses of morphine from 10 to 80 mg/kg for 5 consecutive days.The rats as controls were administered with same volume of normal saline.The expression of Bcl-2 mRNA was assayed with digacin-labelled Bcl-2 cDNA probe by in situ hybridization and the levels of Bcl-2 protein were measured by immunohistochemistry.Results:The evels of Bcl-2 and their mRNA in prefrontal cortex,locus coeruleus,hippocampus and ventral tegmental area significantly decreased as comparied with that in control rats after chronic administration of morphine and naloxone administration.However,Bcl-2 and their mRNA in nucleus accumbens of rats in withdral state were markedly higher than that in nucleus accumbens of morphine dependent rats.Conclusion:Chronic exposure to morphine appears to induce modification of the expression of Bcl-2 mRNA and their protein of rats in the rat brain,which contributes to the development of morphine dependence.

ABSTRACT

BACKGROUND: Multiple applications of opium medicines can induce the accommodative changes of morphology and function in some intracerebral nerve positions. These accommodative changes are important neurobiological bases inducing drug-desire and re-addiction after detoxification. However, the actual molecular mechanism is unclear at present.OBJECTIVE: To investigate the impacts of the generation of heroin-dependence and detoxification on brain-derived neurotrophic factor (BDNF) in rat to provide a laboratorial gist for the participation of BDNS in heroin-dependence and detoxification.DESIGN: A randomized controlled study by employing experimental animals as subjectsSETTING: Mental health center of a medical university affiliated hospital MATERIALS: The study was conducted in the Laboratory of Pharmacology,Faculty of Pharmacology, Chongqing Medical University between March 2004and July 2004. Totally 30 inbreeding clean male SD rats with a bodymass between 200 g and 250 g were obtained from the Experimental Animal Center of the Third Military Medical University of Chinese PLA. Rats were randomly divided into blank control group(control group), heroin-dependent group (heroin group), and naloxone detoxification group(naloxone group) with 10rats each.METHODS: Morphine was subcutaneously injected into the rat with dose-increasing method to establish heroin-dependence rat model. Rats of naloxone group received subcutaneously injection of 2 mg/kg of naloxone to excite abstinent symptoms. The same dose of normal saline (NS) was injected in rats of control group. Model rats of each group were observed biologically and behaviorally. BDNF expression at different brain zone of rats in three different groups was tested with immunohistochemistry and digoxin-labeled oligonucleoide probe in situ hybridization technique.Comparison of the evaluation of abstinent symptoms in rats of each group.RESULTS: In the heroin group, the relative content of BDNF protein was higher in frontal lobe cortex, locus caeruleus and hippocampus than that of the control group( P < 0.05); BDNFmRNA relative content was higher in frontal lobe cortex than that of the control group( P < 0. 05) . In naloxone group, BDNF and its mRNA relative contents in frontal lobe cortex, locus caeruleus and hippocampus were higher than that of heroin group and control group ( P < 0.05 ).CONCLUSION: Chronic administration of heroin could affect BDNF protein and its mRNA expressions in the corresponding brain areas of the rats, which suggests that the change of BDNF expression participates in heroin-dependence and detoxification.
